Laboratory of Molecular Analyses is a formally established research unit of the Institute of Botany of the Polish Academy of Sciences. Its research profile is defined by the activities of the Molecular Biogeography Group. The lab staff is also running the molecular analyses platform open to all institute research units and focused on DNA analyses, and supervising the collection of DNA isolates of the institute.

Main types of analyses carried out in the lab:

  • DNA isolation and purification using various methods, quality check of isolates
  • Amplification of various target DNA regions in Polymerase Chain Reaction (PCR)
  • Restriction site polymorhism analyses in DNA amplicons (PCR-RFLP)
  • Genotyping through Amplified Fragment Length Polymorhism (AFLP) analysis
  • DNA sequencing using Sanger method
  • "Next-generation" sequencing (including amplicon-based metabarcoding and sequence capture-based methods)
  • Single nucleotide polymorphism (SNP) genotyping using RAD-Seq and hyRAD techniques

Laboratory facilities and equipment:

  • DNA isolation site for single sample and 96-plate format, including tissue homogenisers (TissueLyser II, Qiagen/Retsch; Mixer Mill M200, Retsch), high-speed plate centrifuge (Sigma 4-16K) and electronic multidispensing pipettes (Matrix)
  • Electrophoresis site equipped with several power supplies (Amersham Biosciences, BioRad), agarose gel horizontal electrophoresis units (BioRad) and a polyacrylamide gel vertical electrophoresis unit (Amersham Biosciences), a transiluminator and a gel analysis and imaging system (UVITEC Cambridge)
  • PCR site equipped with five thermal cyclers (Life Technologies – Veriti, GeneAmp 9700; Eppendorf – Nexus …)
  • DNA sequencing site with an Applied Biosystems 3500 capillary DNA sequencer and Illumina MiSeq "next-generation" DNA sequencer
  • Several working sites (prePCR, prePCR low DNA content, postPCR) equipped with vortexes (Heidolph Reax Top), desktop centrifuges (Eppendorf MiniSpin), desktop plate centrifuges (Nippon Genetics), Gilson micropipette sets
  • NGS accessory equipment including Qubit fluorometer, Agilent Tape Station 4200, Pippin Prep size fractionator (Sage Science), Covaris ultrasonicator
  • Additional equipment: plate/tube centrifuge with cooling (Eppendorf 5804R), vaccum concentrator (Centrivap, Labconco) laboratory incubator (Binder), shaker with incubation (Heidolph), –20 °C freezers and refrigerators (Liebherr), ice machine (Scotsman), autoclave (Cominox), water destillation and deionisation systems, laboratory scales, magnetic stirrer, water baths

The laboratory additionally supervises the collection of DNA isolates located at the field station in Szarów and assuring long-term storage of DNA samples frozen in –80 °C. The collection has three laboratory deep-freezers (NuAire, VestFrost).
